Accelerated EGM with Simultaneous Dual Nicking & Labeling

This poster presents a new Simultaneous Dual Nicking & Labeling (SNL) workflow for the OhmX™ Platform, designed to accelerate and simplify electronic genome mapping. The SNL chemistry performs both nicking reactions and labeling in a single step and temperature, eliminating multiple setup and heat inactivation steps. The result is a streamlined workflow that reduces total preparation time from two days to less than six hours.

By minimizing high molecular weight DNA breaks and improving tag density, the SNL protocol enables larger contigs, greater genome coverage, and improved structural variant recall compared to the standard sequential workflow. Benchmarking with the Genome in a Bottle dataset demonstrated an 83% increase in contig N50 and a 25% improvement in SV recall while maintaining precision.

Together, these advances establish SNL as a powerful enhancement to OhmX genome preparation, delivering higher-quality assemblies and more complete insight into the structure of the human genome.
